Training for big Stair Climb

I'm training for a stair climb for lung cancer at a hotel in downtown Vancouver. The building is 48 stories tall (490 feet), and there are 739 stairs.

I'm looking for good ways to get more in shape for this event (it's on Feb 20th). Currently I practice by running up my own building from the 1st floor to the 19th where I live daily. At first it was rough, and now I'm capable of jogging it and not passing out after, but going past that would be tough.

Anyone have any good ideas on preparing for this? Currently the first thing that goes is my legs (My body is tired, but not that tired), quads feel dead about 12 stories up and the rest is a bit of a struggle.

i wouldnt cut calories drastically now but dropping a few pounds should make a big diff over 48 stories .
